Transfection-based HR and NHEJ kits use transient transfections to make a pool of cells containing the DNA reporters to allow the investigator to interrogate the 2 major DNA repair pathways. The advantages are:
Our DNA Repair Kits use a novel GFP reporter that converts from GFP negative to GFP positive cells through either NHEJ or HR mechanisms. The plasmids are specifically engineered to follow only one of the two major repair paths found in animal cells. Our Kits are extremely flexible and can be applied in several ways. The Kits in described in this collection and very easy and simply to apply. Most labs work in specific cell line models (Lymphoma, Prostate, Pancreatic cells for example) and are interested in testing DNA repair in those model cells. You can take any cell line of choice and simply introduce two key plasmids simultaneously by transient transfection and analyze the resulting positive cells as a means of DNA repair efficiency. This means that you will be testing a pool of cells that each contain the appropriate plasmid DNA’s. Any cell in the population that does not receive both reporters will be GFP negative and therefore can be ignored. A major advantage of the system is that the investigator can immediately get the system, running and perform experiments with minimal downtime associated with creating stably transfected cell lines.
